Copyright © 2006 Massachusetts Medical Society. n this article, we review recent findings related to estrogen exposure and the risk of breast cancer, the mechanisms that may be involved, and the clinical implications of these findings. The weight of evidence indicates that exposure to estrogen is an important determinant of the risk of breast cancer. The mechanisms of carcinogenesis in the breast caused by estrogen include the metabolism of estrogen to genotoxic, mutagenic metabolites and the stimulation of tissue growth. Together, these processes cause initiation, promotion, and progression of carcinogenesis. Insight into the mechanisms of the causation of cancer by estrogen will identify determinants of susceptibility to breast cancer and new targets for prevention and therapeutic intervention.

Relation between Estrogen and Progesterone Receptor Status with p53, Ki67 and Her-2 Markers in Patients with Breast Cancer
Background: Breast cancer is the most common cancer in women, containing approximately one third of all illnesses in women. Assessment of molecular markers is valuable in predicting the outcome of disease and decision making for optimal treatment. The purpose of this study was to determine the relationship between estrogen and progesterone receptors with Her-2, Ki67, P53, and clinicopathologica...
متن کاملGenetic polymorphisms in the estrogen receptor - α Gene codon 325(CCC}CCG) and risk of breast cancer among Iranian women: a case control study
Abstract Background: The Iranian breast cancer patients are relatively younger than their Western counterparts. Evidence suggests that alterations in estrogen signaling pathways , including estrogen receptor-α (ER- α ), occur during breast cancer development in Caucasians. Epidemiologic studies have revealed that age-incidence patterns of breast cancer in Asians differ from those in Cauca...
متن کاملBioinformatics-Based Prediction of FUT8 as a Therapeutic Target in Estrogen Receptor-Positive Breast Cancer
Abstract Introduction: Estrogen receptor-positive (ER-positive) breast cancer is a subgroup of breast tumors that is more likely to respond to hormone therapy. ER-positive and ER- negative breast cancers tend to show different patterns of metastasis because of different signaling cascade and genes that are activated by estrogen response. Genetic factors can contribute to high rates of metastas...
متن کاملImaging features of estrogen-negative breast cancers: a correlation study with human epidermal growth factor type II overexpression
Background: Estrogen-negative breast cancers have different clinical course, prognostic features and treatment response in comparison to estrogen receptor-positive (ER-positive) breast cancers. Human epidermal growth factor receptor 2 (HER2) oncoprotein has found to have a pivotal role in natural cell growth and cell division and is suggested to be directly related to tumor invasiveness in brea...
متن کاملCyclin D1 enhances the response to estrogen and progesterone by regulating progesterone receptor expression.
Estrogen and progesterone are the defining hormones of normal female development, and both play critical roles in breast carcinogenesis. Cyclin D1 is a breast cancer oncogene whose amplification is linked to poor prognosis in estrogen and progesterone receptor-positive breast cancers. Here we report that cyclin D1 regulates progesterone receptor expression, consequently enhancing responses to e...
